Isolation and Identification of Two Unknown Organisms from a Mixed Culture by means of Selective and Differential Testing Abstract Bacteria make... up an overwhelmingly large portion of the population of life on this planet, and there is so much diversity within the domain itself. It is important to understand how to isolate and identify microorganisms to get a better understanding of the diverse population of bacteria in the world. To find the identity of two unknown species in mixed culture #55, a variety of selective and differential tests were performed. The two species were initially isolated with selective media, and they later were tested on other selective and differential types of media to narrow down the potential identities of the organisms. After experimental observations were taken and analyzed, it was concluded that the Gram-negative microbe in the mixture was Escherichia coli and the Gram-positive bacteria was Enterococcus faecalis.
