Chemistry > Lab Report > University of Great Falls - CHEMISTRY 310Lab 3 Enzyme Kinetics of Tyrosinase (All)
I. Introductions The purpose of this experiment was to observe and analyze the effects of changing mushroom tyrosinase and L-DOPA concentrations on initial reaction rates. Enzyme kinetics is the st... udy of enzyme rates and how these rates are affected by enzyme concentration, substrates, and any inhibitors or activators. The reaction rate is expressed as a change in the concentration of a reactant or product during a given time interval. These rates were calculated and then used to construct Michaelis-Menton and Lineweaver-Burk plots to determine the VMax, KM, and turnover number (Kcat). II. Hypothesis The reaction rate will increase as the volume of enzyme and substrate increases. III. Materials Sodium Phosphate Buffer, 0.1M, pH 7.0 Mushroom Tyrosinase (0.2 mg/mL) L-DOPA, 15 mM Spectrophotometer Stopwatch Cuvettes Test tubes Micropipette IV.
